If I have more than 1 player for lets say Mid and I make 2 strategys, do both play or does only 1 strategy count?

Well, you can only have one strategy for each game type, if you have multiple strategies for one gametype, only one will be used.
Some people in the past have done things where in League they had their "streamers", who are players not being coached, play while in team ranked they used their normal players.
You can't really have it randomly pick for the same queue, but you can use different players for different sides/different queues if you want.
